摘要
以篦子三尖杉的幼茎和嫩叶作为外植体进行组织培养,研究不同激素及不同浓度组合对其诱导愈伤组织的影响,结果表明:篦子三尖杉幼茎比嫩叶的愈伤组织诱导率高;MS+NAA 1 mg.L-1+6-BA 0.15 mg.L-1+2,4-D 1 mg.L-1是诱导幼茎愈伤组织的最佳培养基,诱导率达100%;MS培养基相比B5培养基更适合篦子三尖杉嫩叶愈伤组织的诱导;幼茎在75%的酒精中消毒1 m in,在0.1%的升汞溶液中浸泡15 m in,其消毒效果最佳。
Taken young leaves and stem segments of Cephalotaxus oliveri as explants, the effects of different hormone and proportions on callus induction of Cephalotaxus oliveri were researched. The results indicated the induction rate of young stem callus were higher than that of young leaves callus. The best medium for inducing young stem callus was MS + NAA 1 mg·L^-1 +6-BA 0. 15 mg·L^-1 + 2, 4-D 1 mg·L^-1 and the induction rate reached 100%. MS medium was more suitable for inducing young leaves callus than B5 medium. The best sterilization procedure for young stem was to soak stem for 1 rain in 75% alcohol and 15 min in 0. 1% HgCl2.
