摘要
目的:研究小鼠后肢缺血再灌注对背根神经节(DRG)形态结构的影响。方法:采用暂时阻断小鼠髂总动脉起始处血流的实验模型,随机分为假手术组(A组)、缺血6h再灌注组(B组)和缺血8h再灌注组(C组),观察不同缺血时间再灌注后DRG的超微结构改变。结果:电镜下可见A组为正常细胞结构;B组细胞有局灶性核溶解发生,粗面内质网池扩张但其扩张程度远远低于C组。卫星细胞核孔消失,微血管内皮细胞暗细胞变。C组细胞核仁呈轻度溶解,线粒体变暗,卫星细胞明显增生,可见凋亡小体被卫星细胞吞入,微血管内皮呈暗细胞变,腔内可见崩解小球的空泡。腔隙间隔极度增宽,其间可见坏死细胞。结论:小鼠后肢缺血再灌注可引起DRG超微结构的损伤。
Objective: To explore the effects of dorsal root ganglion (DRG) after hindlimb ischemiareperfusion in mice. Methods: By using the model of occlusion and reperfusion of common iliac arteries of the mice, Male Kunming mice (20-25 g) were randomly divided into 3 groups with different treatments: Sham-treated control group (Group A), ischemia for 6 h reperfusion groups (Groups B) and ischemia for 8 h reperfusion groups (Groups C).The uhrastructure changes of DRG were observed. Results: Under electron microscopes those were observed, Group A: normal cyto-architecture; Groups B: cells had focal karyolysis, the rough endoplasmic reticulum cisterna was less dilated than groups C, nucleopore of satellite cell disap- pea.red, the vas eapiUare endothelial cells became dark cells; Groups C: entoblast light dissolved and chon- driosome became dark, satellite cell had hyperplasy and phagocytosed apoptotic body, vacuole of disaggre- gation bleb were found, laeouna spacing widen and cell necrosis were found. Conclusion: The mice's hindlimb isehemia-reperfusion can cause the ultrastructure damage of the DRG.
