摘要
目的本研究旨在建立一种准确、快速、简便的O139群霍乱弧菌检测方法。方法以荧光纳米颗粒为标记物,采用双抗体夹心法模式制备O139群霍乱弧菌免疫层析检测试纸条。在紫外灯下观察试纸条上质控线和检测线上的荧光信号强度作为结果判定依据。结果用所制备的O139群霍乱弧菌荧光纳米颗粒检测试纸条对9属20种105株菌进行检测,所有20株O139群霍乱弧菌检测结果呈阳性,其余85株非O139群霍乱弧菌均呈阴性反应;用该试纸条检测人工污染的海鱼样品,灵敏性为3.5×104CFU/mL。用自制试纸条和标准法同时检测77份样品(均为水产品),两种方法的总符合率为85.7%。结论O139群霍乱弧菌荧光纳米颗粒检测试纸条的研制成功为O139群霍乱弧菌的快速检测提供了一个极好的检测方法,便于野外检测的开展,具有广阔的应用前景。
Objective To establish a rapid method for the detection of Vibrio cholera O139. Methods Fluorescent nanoparticles were covalently conjugated with monoclonal antibodies against V. cholera O139 and were employed in the development of lateral flow strip assay. The sensitivity, specificity and semi-quantitative of the lateral flow strip were evaluated. Results The sensitivity that detected artificial contaminant fish was 3.5 ×10^4C CFU/mL. The method had good specificity and there were no cross-reaction with non- O139. With this method, semi-quantitative detection of V. cholera O139 can be finished in less than 15 min. Compared with standard method (SN/T1872-2007), the coincidence are 85.7%. Conclusion This lateral flow strip assay is a sensitive, simple, rapid method without the need of special apparatus, so it is of good practical value for the detection of V. cholera O139 in locale.
