摘要
目的观测梗阻性黄疸(OJ)大鼠血浆内皮素(ET-1)及肾皮质微循环的变化,探讨OJ肾损伤的机制及丹参的治疗作用。方法将SD大鼠分为假手术组(SO组);胆总管结扎组(BDL组);丹参干预组(SM组),每组分3、7、10d三个时相,每个时相10只。测定ET-1,血尿素氮(BUN),肌酐(Cr)水平。应用WX-9型微循环显微镜观测3组相应时相肾皮质微血管管径的变化。结果BDL组与SO组比较各时相ET-1水平升高(P<0.05),肾皮质微血管收缩(P<0.05),7、10d血BUN,Cr水平升高(P<0.05)。且随胆道梗阻时间延长变化更加明显(P<0.05)。SM组与BDL组比较各时相ET-1水平下降(P<0.05),7、10d肾皮质微血管扩张,10d血BUN,Cr水平下降(P<0.05)。结论血浆ET-1水平升高及肾脏微循环障碍是梗阻性黄疸肾损害的重要原因,丹参注射液对梗阻性黄疸肾损害有保护作用。
Objective To observe the changes of renal cortex microcirculation, ET-1 in obstructive jaundice models and study the effect of Salvia Miltiorrhiz(SM). Methods 90 SD rats were randomly, divided into three groups: sham operation group(SO) , common bile duct ligation group(BDL) ,Salvia Miltiorrhiz group(SM). The the caliber of renal cortex capillary vessel was measured by the WX-9 microscope, and the levels of ET-1, blood urea nitrogen(BUN) ,creatinine(Cr) at 3,7,10 days were detected. Results As compared with SO group, the ET-1 levels in group BDL increased at every time point, but the caliber of capillary decreased, the levels of BUN and Cr were significantly increased at the 7 and 10 days ( P 〈 0.05 ) , there were more significant changes with the time going( P 〈 0.05 ). AS compared with BDL group, the ET-1 levels in group SM decreased, but the caliber of capillary increased at the 7 and 10 days ( P 〈 0.05 ) , and the levels of BUN and Cr significantly decreased( P 〈 0. 05 ). Conclusion Obstructive jaundice can induce the microcirculation disturbance of renal cortex and increase the ET-1 levels in rats, and Salvia Miltiorrhiz(SM)play an important role in relieving the kidney injury of obstructive jaundice rats.
