MMP-9、OPG在风湿钙化二尖瓣的表达及意义

摘要 目的观察基质金属蛋白酶-9(MMP-9)和骨保护素(OPG)在风湿钙化二尖瓣的表达。方法将手术切取下的二尖瓣共42枚,按病史分为风湿钙化组(24枚)和非风心病组(18枚)。采用SP法进行免疫组化染色观察基质金属蛋白酶-9和骨保护素在2组的表达。结果在风湿钙化组,基质金属蛋白酶-9和骨保护素表达均明显高于非风心病组(P<0.01)。结论风湿瓣膜的钙化与基质金属蛋白酶-9所引起的细胞外基质降解和重塑以及骨保护素所诱导的成骨样骨形成密切相关。 Objective To observe the expression and significance of matrix metalloproteinase-9 and osteoprotegerin in calcified mitral valves of rheumatic heart disease. Methods 42 mitral valves cut by operation were divided into calcified rheumatic group and non-calcified rheumatic group, and the expression of matrix metalloproteinase-9 and osteoprotegenn was observed by immunohistochemistry method. Results The expression of matrix metalloproteinase-9 and osteoprotegerin in calcified rheumatic group significantly increased than that in non calcified rheumatic group( P 〈 0.01). Conclusion Both matrix metalloproteinase-9 and osteoproteger are highly expressed in calcified rheumatic group, which proves that calcify of rheumatic mitral valves is related with the degradation and remodeling of extra cellular matrix by matrix metalloproteinase-9, and closes with osteoblastlike bone formation by osteoprotegerin.
