摘要
对常用4种双歧杆菌培养基进行比选,确定出产双歧杆菌高的基础培养基,采用正交试验通过改变碳源、添加低聚果糖及pH值的内源调节优化基础培养基,优化培养基为在比选出的基础培养基中加0.5%低聚果糖和1%碳酸钙。利用优化培养基培养双歧杆菌,当培养液pH值低于5.5时,用6mol/LNaOH调节pH值为6.0~6.5,在厌氧条件下37.5℃培养24h,活菌数达到1.7×1010cfu/mL。
The optimal medium of Bifidobacterium was selected from the 4 base medium and the base media were furthere optimized through orthogonal experiment. 0.5% fructooligosaccharide and 1% calcium carbonate were added into the base medium, pH was adjusted and controlled between 6.0-6.5 using 6 mol/L NaOH when pH was lower than 5.5. The viable cell number of Bifidobaicterium could reach 1.7×10^10cfu/ml in the optimal growth medium at 37.5℃ for 24 h under anaerobic condition.
