摘要
目的探讨针刺对家兔实验性动脉血管平滑肌细胞c-myc及c-foc基因表达的影响。方法家兔随机分为正常对照组(8只),再狭窄组(24只)。再狭窄组应用PTCA球囊导管对实验兔行双侧髂动脉内膜剥脱以建立动脉粥样硬化模型,术后1月髂动脉造影示50%以上狭窄者再行PTCA球囊扩张术,以建立再狭窄模型成功后,然后将造模动物随机分成模型组、针刺组、普罗布考组,每组8只。自术前2d~术后14d为1疗程,针刺内关(双)、厥阴俞(双)、心俞(双)、膻中、足三里(双)等穴。标本石蜡切片,免疫组化,DAB显色,苏木素复染。观察各组血管平滑肌细胞c-myc、c-foc基因表达。结果模型组平滑肌细胞c-myc及c-foc基因表达明显高于正常组(P<0.01);针刺组c-myc及c-foc基因表达低于模型组(P<0.01)。结论针刺能抑制再狭窄兔血管平滑肌细胞增殖,抑制c-myc及c-foc基因表达,调节平滑肌凋亡,可能是针刺预防动脉再狭窄的作用机制之一。
Objective To explore the effect of acupuncture on treating vascular smooth muscle cell and gene expression of c-myc and c-foc in rabbits. Methods The rabbits were randomly divided into normal control group (n=8) and re-stricture group (n=24). To establish atherosclerosis model applying PTCA sacculus proprius to denude arteria iliaca inner membrane two sides in re-stricture group. One month after operation,PTCA sacculus proprius dilatation was made if arteria iliaca opacification indicated that there were artery stricture above 50 percent,after succeeding in establishing the re-stricture models,then the model animals were divided into model,acupuncture and probucol groups (each n=8). It was a course treatment form the first two days before operation to the 14th day after operation,acupuncturing acupoints of Neiguan (pair),Jueyinshu (pair),Xinshu (pair),Danzhong,Zusanli (pair) and so on. Paraffin section,immunohistochemical method,DAB coloration,and hematoxylin staining were used,and then the c-myc and c-foc gene expression of smooth muscle cell in each group was observed. Results The c-myc and c-foc gene expression of smooth muscle cell in model group was prior to that in control group significantly (P0.01). The gene expression of c-myc and c-foc in acupuncture group was low than that in model group (P0.01). Conclusions Acupuncture can restrain the hyperplasy of restenosis of vascular smooth muscle in the rabbits,restrain the gene expression of c-myc and c-foc and adjust the apoptosis of smooth muscle. This may be one of the mechanisms of preventing arterial restenosis with acupuncture.
