摘要
5-氟尿嘧啶与氢氧化钾作用得到5-氟尿嘧啶单钾盐,5-氟尿嘧啶单钾盐与氯乙酰乙酯得到N1-乙酸乙酯取代5-氟尿嘧啶,再经酸性条件下水解合成了5-氟尿嘧啶-1-基乙酸,产率85%,产品纯度99.52%,并采用红外、核磁、紫外的方法表征了产物的结构,确定了产品的最佳反应条件。采用此法制备5-氟尿嘧啶-1-基乙酸原料便宜易得,操作方法简单,产物具有较高的产率和纯度。
5 - fluorouracil potassium salt was synthesized by the reaction of 5 - fluorouracil and potassium hydroxy, and 5 - fluorouracil potassium salt reacted with ethyl chloroaceticate, then hydrolyze in HC1, obtained 5 - fluorouracil - 1 -yl with the yield of 85% and 99.52% purity(by HPLC). FT- IR, 1H -NMR and UV were used to characterize the structure of the product, and the optimum reaction condition was confirmed. This synthetic method had the merits such as cheapness of materials, simplification of manipulation, and high in both yield and purity.
