雌激素对去卵巢EAE大鼠IFN-γ影响的相关性研究

摘要 目的探讨EAE的发病机制和应用雌激素治疗EAE的作用机制,为EAE的免疫病理机制提供新的理论支持,也为今后临床应用雌激素辅助治疗EAE自身免疫性疾病提供实验依据。方法制备去卵巢后的EAE动物模型,随机分组。观察各组在不同时间点的血清E2浓度及血清和脑脊液中白蛋白含量比值,观察体重变化、发病率等临床指标。处死动物,取脑和脊髓用免疫组化技术检测CNS中不同部位干扰素-γ(interferon-γ,IFN-γ)的表达,并对其进行相关分析。结果IFN-γ主要由进入CNS的淋巴细胞产生,免疫组化染色阳性表达表现为血管渗出的淋巴细胞周围有不同程度着色,且表达程度与病情轻重相一致。结论EAE早期CNS中已有少量IFN-γ表达,活动期表达增加,病情减退表达随之下降。
