摘要
通过FX-4000柔性基底加载系统对人舌鳞癌Tca8113细胞进行力学环境下的培养、流式细胞仪检测分析,研究力学刺激对Tca8113细胞增殖的影响。实验结果显示:细胞在不同力学刺激下(拉伸波形分别为方波、正弦波、三角波,拉伸幅度分别为5%、10%,频率为0.5 Hz,加载时间4 h),与静态对照组比较,方波组细胞的增殖速率得到明显的抑制,且方波10%组细胞的增殖速率明显小于方波5%组细胞的增殖速率;与静态对照组比较,正弦波5%组、10%组、三角波5%组的增殖速率无明显差异,三角波10%组细胞的增殖速率得到明显的抑制。研究表明:拉伸力学刺激的波形和幅度是影响Tca8113细胞增殖行为的重要因素。
The human squamous carcinoma of tongue cell line Tca8113 cells were cultured on six-well culture plates under the mechanical environment by FX-4000 strain system,the effects of stretch on proliferation of Tca8113 cell were examined and analyzed by flow cytometry.Experimental regimen: square wave,sine wave and triangle wave with 5% and 10% elongation amplitude,4h duration,and frequency of 0.5Hz.Experimental results: The cellular proliferation rate was reduced significantly in square wave group and triangle wave(10% elongation) group compared with the control group and decreased with the stretch amplitude;There was no significant difference in proliferation rate between sine and triangle wave(5% elongation) group and control group.It is concluded that waveform and elongation amplitude of cyclic stretch play important role in the proliferation of Tca8113 cells.
