摘要
目的探讨在散发性乳腺癌中影响易感基因(BRCA1)蛋白表达的因素及其与临床指征的关系。方法运用免疫组织化学SP法检测10例乳腺导管内乳头状瘤和48例乳腺癌组织中BRCA1的表达。结果在10例乳腺导管内乳头状瘤病例中,BRCA1蛋白阳性位于细胞核,阳性表达率为100%(10/10);48例乳腺癌组织中BRCA1蛋白阳性表达位于细胞浆/细胞核和细胞浆,阳性率为56.3%(27/48),二者差异有统计学意义(P<0.01);BRCA1的失表达率在>40岁的发病年龄组高于≤40岁的发病年龄组(P<0.05)。结论在散发性乳腺癌中,BRCA1的失表达高于良性肿瘤,并与高发病年龄相关,BRCA1基因表型的改变为乳腺癌的预后判断提供了新依据。
Objective To investigate the expression of BRCA1 protein in sporadic breast cancer. Methods The expression level of BRCA1 protein was detected by immunohistochemistry(IHC)with Streptavidin-Peroxidase(SP) method in benign breast tissues and sporadic breast carcinoma tissues. Results The positive expression rates of BRCA1 were 100 % (10/10)and 56.3 % (27/48)in benign breast tissues and sporadic breast carcinoma tissues, respectively. The difference in BRCA1 expression between the two groups was significant(P〈0.01 ). In the sporadic breast cancer tissues,BRCA1 express in cytoplasm or in cytoplasm and nuclei. Furthermore,loss of BRCA1 expression in breast cancer tissues was correlated with the age of the patients. Conclusion Loss of BRCA1 expression is higher in sporadic breast cancer than in benign breast tissues;Loss of BRCA1 expression in sporadic breast is correlated with age. Altered BRCA1 phenotype may therefore provide an additional prognostic parameter for breast cancer.
