摘要
目的通过实验对正常乳腺组织和浸润性乳腺癌组织中淋巴增强因子-1(LEF-1)转录因子的蛋白质表达水平进行分析,观察其差异。方法对从手术获得的新鲜乳腺组织做冰冻切片,经H&E染色分析鉴定出正常乳腺组织和乳腺浸润癌组织。用显微切割方法获取目标组织,再采用蛋白质免疫印迹法(Westen blot)检测正常乳腺组织和乳腺浸润癌组织中LEF-1转录因子的蛋白质表达差异。结果乳腺浸润癌组织中的LEF-1转录因子的蛋白质表达水平高于正常乳腺组织中的LEF-1转录因子的蛋白质表达水平。结论LEF-1转录因子的表达与乳腺癌的浸润相关,正常乳腺组织和乳腺浸润癌组织中的LEF-1转录因子的蛋白质表达水平的分析是研究LEF-1转录因子的调控作用机理的基础。
Objective This study analyse the level of protein expression of LEF- 1 transcription factor in normal breast tissue and invasive breast cancer tissue. Methods Fresh pathology material from surgical operation were performed frozeen dissecting, and then stain by H&E staining method. Examining the slice and divided tussues into two groups-normal breast tissue and invasive breast can- cer. To examine the difference of protein expression level of LEF-1 transcription factor in the normal mammary gland tissue and inva- sive breast cancer tissue by Western blot method. Results The level of protein expression of LEF- 1 transcription factor in the invasive breast cancer tissue was higher than LEF-1 transcription factor in the normal mammary gland tissue. Condusion Expression of LEF- 1 transcription factor are interrelated with infiltration of breast cancer. This research is a basis for further study of the regulatory mech- anism of LEF-1 transcription factor in the invasive breast cancer.
