摘要
通过对以往的叶绿体DNA提取方法进行比较研究,建立了一个快速、简便提取栓皮栎叶绿体DNA的方法,其主要步骤包括叶绿体分离、DNA酶处理、去蛋白和纯化。结果表明,这种方法对叶绿体DNA的提取具有高纯度、高质量和快速简便等优点。同时对获得的叶绿体DNA进行了RAPD分析,并摸索出其最适反应条件。
A fast, simple and convenient method is established to isolate and characterize the cpDNA in leaves of Quercus variabilis.The protocol includes: chloplast isolation, deoxyribonuclease treatment, deproteinisation and purification of the DNA.The technique is inexpensive and appropriate for the isolation of cpDNA and is easy to obtain cpDNA with high purity and high quality. RAPD analysis system has also been established for study of this cpDNA.
