甘草提取物防治变应性鼻炎的实验研究

Experimental study of glycyrrhiza water extractives in treating guinea pigs with allergic rhinitis

目的:验证甘草提取物治疗变应性鼻炎是否有效。方法:建立变应性鼻炎的豚鼠模型,通过用甘草提取物滴鼻治疗后来检测鼻分泌物、鼻粘膜中主要致病物质的变化情况。包括HE染色和Chromotrope 2R特染分别检测豚鼠鼻分泌物及鼻粘膜组织中炎性细胞总数及嗜酸性粒细胞数;TUNEL法检测鼻粘膜组织中嗜酸性粒细胞凋亡;甲苯胺蓝染色检测鼻粘膜组织中肥大细胞数及脱颗粒数;ELISA法检测鼻粘膜组织中组胺含量。结果:研究发现用甘草提取物滴鼻治疗后变应性鼻炎豚鼠的鼻痒、喷嚏、流涕等症状得到明显减轻。检测发现甘草提取物滴鼻治疗后变应性鼻炎豚鼠鼻分泌物及鼻粘膜组织中的炎性细胞总数、嗜酸性粒细胞数明显减少(P(0.05);豚鼠鼻粘膜组织中的嗜酸性细胞凋亡率明显增加(P(0.05);肥大细胞数及脱颗粒数明显减少(P(0.05);组胺含量明显减少(P(0.05)。其中甘草提取物高剂量组鼻分泌物及鼻粘膜组织中的这些指标的减少程度与地塞米松组相当。结论:从动物实验研究结果证明甘草提取物在治疗变应性鼻炎上是有效的。

Objective To investigate the therapeutic effect of glycyrrhiza water extractives on the animal model of guinea pigs with allergic rhinitis. Methods The animal model of guinea pigs with allergic rhinitis set upby instilling toluence 2, 4 diisocyanate （TDI） as the sensitizer were treated with glycyrrhiza water extractives. At the end of the treatment, the differences in the behavior science were observed ; the levels of total inflammatory cells and eosinophils in nasal lavage fluid and nasal mucosa were studied by HE stain and Chromotrope 2R stain; and the level of eosinophils apoptosis was determined by TUNEL test; and the level of mast ceils and degranulation was determined by toluidine blue stain ; at last the level of histamine was determined by ELISA. Results The behavior science score of the glycyrrhiza water extractives treatment group was lower than that of the control ; the levels of total inflammatory cells and eosinophils in nasal lavage fluid and nasal mucosa were lower than that of the control （ p 〈 0. 05） ; and the level of eosinophils apoptosis was higher than that of the control （p 〈 0. 05） ; and the level of mast cells and degranulation was lower than that of the control （ p 〈 0. 05） ; and the levels of histamine were lower than that of the control （ p 〈 0. 05）. At last the decreased levels of these pathogenic factors of high - dose glycyrrhiza treatment group was similar with dexamethasone treatment group. Conclusion Glycyrrhiza water extractives has certain effects in treating allergic rhinitis through blocking multi - links of AR.
