罗格列酮对高糖状态下人肾小管上皮细胞转化生长因子β_1表达的影响 被引量:1

摘要 目的研究罗格列酮对高糖状态下人肾小管上皮细胞转化生长因子β1(TGF-β1)表达及细胞活力的影响。方法体外培养人肾小管上皮细胞(HK-2),首先观察高糖状态下HK-2细胞TGF-β1表达情况,然后用不同浓度的罗格列酮刺激48h及使用相同浓度的罗格列酮刺激不同时间,利用Westernblotting法观察各实验组细胞TGF-β1蛋白表达情况,使用ELISA法观察各实验组培养液中Ⅳ型胶原表达的变化。结果罗格列酮可呈时间和浓度依赖性抑制高糖状态下人肾小管上皮细胞TGF-β1、Ⅳ型胶原酶的表达(P<0.05)。结论罗格列酮可以保护高糖状态下肾小管上皮细胞,部分机制是通过抑制TGF-β1、Ⅳ型胶原酶的表达。
