生理药动学模型预测二甲双胍在人体内的动力学过程 被引量:1

摘要 目的:建立二甲双胍在人体内的血流限速生理药动学模型,预测正常和病理条件下,药物在人体内的动力学过程。方法:应用ACSLXtreme与3P97软件进行求解模拟,并将模拟值和文献值比较分析。结果:模型模拟二甲双胍的药动学参数为:Cmax1.18μg/mL,tmax2.33h,t1/2a2.84h,AUC8.08μg/(mL·h-)与文献报道值基本吻合。结论:生理模1型能够准确预测二甲双胍在机体内的动力学过程。 Objective: To develop a flow-limited, physiology based pharmacokinetie (PBPK) model for predicting kinetica process of Metformin in the body trader healthy and pathological conditions. Methods: The simulation equations were solved by the ACSLX treme software and the simulated concenlrations were compared with the measaued ones. Results: Simulated pharmacokinetic parameters of Mefformin were : Cmax 1.18μg/mL, tmax 2.33 h, t1/2a, 2.84 h, AUC 8.08μg/(h·mL-1) and these parameters were according to the reported values. Conclusion: PBPK can accurately predict the kinetica process of the drug in the body.
