雷抑素对肝移植大鼠肝Kupffer细胞的影响 被引量:1

EFFECTS OF LEFLUNOMIDE PRETREATMENT ON KUPFFER CELLS IN RAT LIVER TRANSPLANTATION

摘要 利用大鼠原位肝移植模型,观察移植术前应用雷抑素(Lef)对大鼠肝Kupffer细胞形态及功能的影响。结果显示,对照组肝移植术后3小时血清肿瘤坏死因子(TNF)、ALT活性显著升高,肝组织丙二醛(MDA)水平也显著增加;用药组血清TNF、ALT活性及肝组织MDA水平显著下降。电镜检查,对照组肝Kupffer细胞呈典型“活化”表现,而用药组肝Kupffer则呈非“活化”表现。结果提示,移植术前应用Lef可以抑制TNF及氧自由基的产生,Lef通过抑制肝Kupffer细胞的“活化”而发挥减轻肝冷缺血再灌注损伤作用。 Rat orthotopic liver transplantation model was used to determine the effects of Leflunomide pretreatment on thestructure and function of Kupffer cells. The results show that the levels of TNF and ALT in blood,and MDA in liver tissue increased rapidly 3 hours after transplantation in controls. The Leflunomide - pretreated recipients had lower levels of TNF andALT,and the level of MDA decreased significantly. The Kupffer cells from controls were activated in appearance,while the onesfrom groups preteated with Leflunomide showed little evidence of activation by electron microscopy. The data indicate that freeradicals and TNF production are suppressed by Leflunomide pretreatment. Through its suppressive effect on Kpffer cells,leflunomide may help minimize cold hepatic ischemic reperfusion injury.
