摘要
目的探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)联合低剂量顺铂(DDP)对食管癌EC9706细胞增殖的影响。方法单用DDP和单用TRAIL作用于EC9706细胞24、48、72 h后,利用MTT法检测各处理因素对EC9706细胞增殖的影响;TRAIL联合低剂量DDP作用48 h后,利用MTT法检测各处理因素对EC9706细胞增殖的影响。结果随着单用DDP和单用TRAIL浓度的提高,作用时间的延长,其对EC9706细胞的抑制作用增强。TRAIL与低剂量DDP联用可明显抑制EC9706细胞的增殖,与单用DDP或单用TRAIL相比,细胞存活率显著降低;联合DDP组和DDP+TRAIL组细胞增殖率均随着DDP浓度的增加而下降。结论TRAIL与低剂量DDP联合应用,可以抑制食管癌EC9706细胞的增殖,TRAIL能增强食管癌细胞对DDP的敏感性。
Objective To investigate the effect of TRAIL in combination with low dosage DDP on proliferation of esophageal cancer EC9706 cells.Methods TRAIL and DDP were used to EC9706 for 24 h,48 h and 72 h respectively.Then cell viability was detected by MTT.TRAIL in combination with low dosage DDP were used to EC9706 for 48 h.Then cell viability was evaluated also by MTT.Results With the increasing dosage of TRAIL or DDP,the proliferations of the cells were decreased correspondly.After the treatment with TRAIL plus low dosage DDP,the EC9706 cell growth was inhibited drastically compared with TRAIL alone and DDP alone.Proliferations of cells both in the combination DDP group and the DDP+TRAIL group were inhibited in a concentration-dependent manner.Conclusion TRAIL plus low dosage DDP can inhibit proliferation of esophageal cancer EC9706 cells,TRAIL could increase the sensitivity of the EC9706 cells to DDP.
