摘要
目的探讨荷人乳腺癌裸鼠移植模型的病理学特点及生物学性状,为乳腺癌病因学、发病学研究提供可靠的工具。方法在裸鼠右侧腋窝注射人乳腺癌MCF-7细胞悬液0.1mL(5×108个/mL),建立乳腺癌裸鼠动物模型。混合干预组小鼠,肿瘤接种部位旁皮下注射瘦素0.2mL(15×104ng/mL)和雌激素0.2mL(0.15mg/mL),每天1次;雌激素组注射雌激素0.2mL(0.15mg/mL);瘦素组注射瘦素0.2mL(15×104ng/mL);空白组注射0.9%氯化钠溶液,每日观察肿块生长情况及肿瘤体积变化。宰杀取材,观察肿瘤组织学特征。结果(1)空白组肿瘤移植成功率为33.3%(10/30),瘦素组为46.7%(14/30),模型失败。混合干预组肿瘤移植成功率为96.7%(29/30),雌激素组为93.3%(28/30),两组间无明显差异(P〉0.05);(2)混合干预组裸鼠平均肿瘤最长径明显优于雌激素组,差异具有统计学意义(P〈0.05);混合干预组裸鼠平均体积明显优于雌激素组,差异具有统计学意义(P〈0.05);(3)病理诊断符合浸润性导管癌。结论(1)MCF-7裸鼠模型的成功建立,需体外补充雌激素。(2)瘦素有增强雌激素刺激MCF-7细胞在裸鼠体内增生的作用;(3)移植的肿瘤细胞保持了人乳腺癌肿瘤细胞的部分病理学特点及生物学特性,可以为病因学、发病学、临床治疗及疗效判断提供可靠的工具。
Objective We established a model of human breast cancer in nude mice, to discuss the fea- ture of pathology and biology of breast cancer, and give help to establish tools of pathogen research. Methods Human breast cancer cells MCF-7 were subcutaneously injected into the right armpit of nude mice to establish human breast cancer models. The mice were divided into composite group, estrogen group, leptin group and the blank group (30 in each). In the composite group, estrogen and leptin were injected into peripheral region of the tumor daily. In the estrogen group, estrogen was injected. In the leptin group, leptin was injec- ted. In the blank group, physiological saline was injected. Tumor growth was observed, and the volume of the tumor was recorded. The tumor tissues obtained from the mice were pathologically examined. Results ( 1 ) The tumor-taking rate of leptin group and the blank group were 46.7 % (14/30) and33.3 % ( 10/30), and the mode is failure. In composite group and estrogen group they were 96.7% (29/30) and 70% (28/30). There was not significant difference between them ( P 〈 0.05 ). (2) The differences of average diameter and volume were statistically significant between the composite group and the estrogen group( P 〈 O. 05 ). (3)Pa- thology diagnosis was invasive ductal carcinoma. Conclusions ( I ) Establishing human breast cancer mod- el in nude mice need the stimulation of estrogen. The tumor-taking rate of nude mice has no relationship with leptin. (2) In the study in vivo, leptin as same as estrogen has stimulating effect on MCF-7 cells proliferation. (3) The transplanted cancer cell partly have the pathology and biology features of the human breast cancer cells and give help to establish tools of pathogen research.
