二甲双胍对胃癌细胞株增生及CyclinD1表达的影响 被引量:5

Effect of metformin on the expression of Cyclin D1 in human gastric cancer cells

摘要 目的观察二甲双胍在体外对人胃癌细胞株SGC-7901生长的作用,并初步探讨其作用机制。方法体外培养胃癌SGC-7901细胞,MTT法测二甲双胍在不同浓度和不同作用时间对胃癌细胞增生的影响;Westernblot法检测二甲双胍对胃癌细胞CyclinD1表达的影响。结果MTT法结果显示:用不同浓度(50mmol/L、100mmol/L)的二甲双胍处理胃癌细胞SGC-7901在24h、48h、72h后,50mmol/L二甲双胍对于胃癌细胞生长抑制率分别为32.93%、48.64%和61.40%,100mmol/L二甲双胍对于胃癌细胞生长抑制率分别为35.34%、75.4J4%和88.30%,不同浓度的二甲双胍对于胃癌细胞生长抑制率与对照组相比具有显著性差异(P〈0.05),100mmol/L二甲双胍对于胃癌细胞生长抑制与50mmol/L二甲双胍对于胃癌细胞生长抑制率相比具有显著性差异(P〈0.05)二甲双胍呈时间、浓度依赖性抑制胃癌细胞的增生。胃癌细胞高表达CyclinD1,Westernblot检测表明二甲双胍能显著降低CyclinD1蛋白的表达,且呈一定时间、剂量依赖性。结论二甲双胍可以下调胃癌细胞株SGC-7901CyclinD1的表达,抑制胃癌细胞的增生。 Objective To investigate the effect of metformin on human gastric cancer line SGC-7901 in vitro, trying to explore the mechanism involved. Methods Human gastric cancer SGC-7901 cell was cul- tured in vitro, MTT was used to study the effects of metformin on cell growth with different concentration or different time. Western blot was used to investigate the influence of mefformin on the expression of Cyclin D1 in cell line . Results The result of MTT showed human gastric cancer SGC-7901 cell was cultured in vitro and was exposed to metformin in different concentration (50 mmol/L, 100 mmol/L) for 24 h,48 h,72 h. The inhibitory rates of 50 mmol/L metformin on the effects of metformin on cell growth was 32.93%, 48.64% and 61.40% and the inhibitory rates of 100 mmol/L metformin on the effects of metformin on ce]~l growth was 35.34%, 75.44% and 88.30%. The proliferation of SGC-7901 cell was inhibited by different concentration (50 mmol/L, 100 mmol/L) metformin and , the inhibitory rates were significantly decreasing compared with the control group( P 〈 0.05 ). The inhibitory rate treated with 100 mmol/L metformin was significandy decreasing compared with 50 mmoL/L metformin group( P 〈 0.05 ). The inhibitory rate treated with metformin at different time (24 h ,48 h, 72 h) was significantly different( P 〈 0.05 ). M etformin could inhib- it the proliferation of SGC-7901 cells, with dose and time-related effects. Cyclin D1 expressed in high level in SGC-7901 and metformin significantly decreased the expression of Cyclin D1, with dose and time-related effects. Conclusion metformin could inhibit the proliferation and lower the expression of Cyclin D1 in human gastric cancer line SGC-7901 in vitro.
