实验性自身免疫性重症肌无力大鼠胸腺caspase 9和caspase 3的表达 被引量:3

Expression and significance of caspase 9 and caspase 3 in the thymus of experimental autoimmune myasthenia gravis rats

摘要 目的探讨凋亡相关基因caspase 9和caspase 3在实验性自身免疫性重症肌无力(EAMG)大鼠胸腺中的表达及其意义。方法采用TUNEL法检测胸腺凋亡细胞;用免疫组化法检测正常对照组大鼠和EAMG组大鼠胸腺中caspase 9和caspase 3蛋白表达。结果正常对照组和EAMG组大鼠胸腺中细胞凋亡指数(IA)分别为(38.70±8.82)%,(27.73±5.76)%;EAMG组大鼠胸腺中caspase 9和caspase 3的表达明显下降,较正常对照组比较差异有统计学意义,且胸腺中caspase 9和caspase 3的表达与细胞IA在正常对照组和EAMG组分别呈显著正相关。结论胸腺细胞凋亡障碍可能是EAMG的发病机制之一;caspase 9和caspase 3蛋白表达在介导EAMG大鼠胸腺细胞凋亡障碍的过程中可能具有重要作用。 Objective To investigate the expression and effect of apoptosis-related gene caspase 9 and caspase 3 in experimental autoimmune myasthenia gravis(EAMG)rats.Methods Apoptosis thymocytes were detected by TUNEL method;Expressions of caspase 9 and caspase 3 were detected in thymus of the normal rats and the EAMG rats by the immuohistochemical method.Results The apoptosis index(IA)was(38.70±8.82)%,(27.73±5.76)% in the thymus of the normal rats and the EAMG rats,respectively.The expression of caspase 9 and caspase 3 in the thymocytes were down-regulated in the EAMG rats,and there was significant difference between the normal rats and the EAMG rats.Moreover the expressions of caspase 9 and caspase 3 were positively related to IA in the both groups.Conclusion The abnormity of apoptosis thymocytes maybe one of the mecheanisms of EAMG,caspase 9 and caspase 3 play an important role in this process of EAMG.
