HSCs移植预处理对骨髓微环境损伤的研究

The injury of irradiation preconditioning for allo-HSCT on bone microenvironment

摘要 目的研究异基因造血干细胞(HSCs)移植预处理-清髓照射对受者骨髓微环境的损伤。方法20只BALB/C小鼠随机分为:正常对照组(10只);单纯照射组(10只),予8.5Gy照射,剂量率0.57Gy,距离2.33m。显微镜下计数外周血中白细胞的变化,流式细胞术分析外周血中内皮细胞(ECs)的变化;病理和电镜观察骨髓微环境的损伤。结果照射后外周血中白细胞减少,ECs升高,第5天达高峰,持续至第7天,与正常组相比,差异有统计学意义(P<0.05)。照射后3d光镜下观察骨髓微环境中大量出血;电镜下ECs和基底膜间被损坏。结论移植预处理中的清髓照射可引起严重的骨髓微环境中内皮损伤。 Objective To investigate the injury of bone microenvironment in allo-HSCT preconditioning-irradiation.Methods Twenty BALB/C mice were separated randomly into two groups,the control group and irradiation group.Mice subjected to irradiation of 8.5 Gy at rate of 0.57 Gy/min at a distance of 2.33 m.Peripheral blood was collected to assay the white blood cells and the circulating endothelial cells(ECs)by inverted phase contrast microscope and flow cytometry.The histological and electron microscope analysis of tissues were collected at the same time.Results The white blood cells decreased after irradiation;in irradiation group the numbers of ECs were elevated after conditioning,then up to the peak at day 5(P0.05),and kept the high level to the 7th days.After 3 day intramedullary hemorrhage was detected by light microscopy.ECs and the basement membrane were destroyed by TEM.Conclusion Irradiation can induce ECs injury of bone microenvironment in allo-HSCT preconditioning.
