A study on metabonomic changes of human renal carcinoma biopsy tissues using high-resolution magic-angle spinning ~1H magnetic resonance spectroscopy

摘要 目的应用高分辨魔角旋转核磁共振1H谱研究肾脏肿瘤组织的代谢变化。方法选取肾癌肿瘤组织和癌旁对比组织标本,分别作病理分析和高分辨魔角旋转核磁共振1H谱检测。结果通过对比发现乳酸、胆碱类物质(胆碱、磷酸胆碱和甘油磷酸胆碱)及肌醇在肾癌组织中明显升高。结论肾癌组织的代谢特征明显不同于癌旁对比组织,这可以为肾癌的诊断提供分子水平上的代谢依据。 Objective To study the metabonomic changes of human renal carcinoma biopsy tissues using high-resolution magic-angle spinning 1H magnetic resonance spectroscopy(MRS).Methods The metabolic profiles in renal tumors and adjacent non-involved renal tissues were measured by high-resolution magic-angle spinning 1H MRS.The tissue samples were stained with the routine haematoxylin and eosin method for histopathological assessment.Results The metabonomic analysis showed that the renal carcinoma tissues contained higher levels of lactate,total cholines and myo-inositol as compared to the non-involved renal tissues.Conclusion The results of this study suggest that the metabonomic profiles of renal tumors are significantly different from those of non-involved tissues,which may be used as references at molecular levels for the diagnosis of renal carcinoma.
