叠氮胸苷诱导人类慢性粒细胞白血病细胞株K562凋亡的研究 被引量:4

摘要 目的观察叠氮胸苷(AZT)对人类慢性粒细胞白血病细胞株K562增殖的抑制作用,探讨其治疗白血病的可行性。方法以不同浓度AZT处理K562细胞,用四氮甲唑蓝实验测定AZT作用于细胞株48 h的IC50,瑞氏染色观察细胞凋亡形态学变化,流式细胞仪检测细胞周期变化,Annexin-V/PI双染法分析细胞凋亡比例。结果AZT对K562细胞株有明显增殖抑制作用,且抑制作用呈时间与浓度依赖性增长;AZT处理48 h后可见细胞凋亡形态学变化及白血病细胞株S期比例增大;Annexin V/PI双染法显示早期凋亡细胞比例增大。结论AZT有诱导白血病细胞凋亡的作用。 Objective To observe the inhibitory action of AZT on leukemia cell strain K562,and explore the feasibility to treat leukemia.Methods K562 cells were treated with different doses of AZT.After 48 h,cytotoxicity was determined by the MTT assay.The Morphological changes of apoptosis were observed by Wright's staining.Cell cycle and apoptosis rate were analyzed by Annexin-V/PI doubling staining flow cytometry assay.Results AZT could inhibit the growth of K562 cells obviously in a dose-dependent and time-dependent fashion.When treated with AZT for 48 h,K562 cells were observed typical apoptosis change by Wright's staining,and flow cytometry revealed both the S phase rate and apoptosis rate of the leukemia cell increased.Conclusion AZT could induce the apoptosis of K562 cells.
