变异链球菌荧光报告株与野生菌株生长能力及生物膜形成能力的比较 被引量:4

摘要 目的:评价变异链球菌荧光报告株(UA140-mrfp)与野生菌株(UA140)生长能力、生物膜形成能力的差异。方法:测定报告株与野生菌株生长曲线,生物膜形成量,扫描电镜观察不同时间段生物膜结构,利用SPSS10.0统计软件进行重复测量设计方差分析。结果:报告株与野生菌株的生长曲线可明显看出3个阶段,均是在0~2h为迟缓期,2~6h为对数生长期,6h以后为稳定期;同期各时间点报告株相比野生菌株A600值略低,但无明显差别。组间比较发现两菌株生物膜形成量无统计学意义(P>0.05)。不同时间段生物膜结构相近,高倍镜下细菌形态无改变。结论:报告株与野生菌株生长能力,生物膜形成能力相近,可用报告株代替野生菌株在激光共聚焦显微镜下研究变异链球菌的生物膜结构。 AIM:To compare the growth and biofilm forming capability of a wild-type Streptococcus mutans strain(UA140) and a fluorescence protein reporter gene-labeled Streptococcus mutans strain(UA140-mrfp).METHODS:The growth of UA140 and UA140-mrfp in BHI broth were measured at different time points at OD600.Biofilm amount and structure were observed with scanning electron microscope(SEM).Data were analyzed by "repeated measurement design ANOVA" with SPSS 10.0.RESULTS:There was no significant growth difference between the 2 strains although the OD 600 value of UA140-mrfp was slightly lower than that of the UA140(P〉0.05).SEM demonstrated that there was no significant difference on the amount of biofilm between the two groups(P〉0.05).Furthermore,the structure of the biofilms was similar.CONCLUSION:UA140-mrfp could be used as a substitute for UA140 in the research of biofilm formationce in vitro model systems.
