摘要 目的研究高糖对人腹膜间皮细胞间隙连接蛋白(connexin43)表达的影响。方法分离培养人腹膜间皮细胞,细胞分为3组:正常糖组(含5.5 mmol/L葡萄糖)、高糖组(含140 mmol/L葡萄糖)和渗透压对照组(含5.5 mmol/L葡萄糖加24.5 mmol/L甘露醇),培养24、48 h后,应用Northern blot、Western blot方法检测con-nexin43 mRNA和蛋白质表达,比较3组之间的差异。结果正常糖组腹膜间皮细胞表达丰富的connexin43,高糖环境下培养的腹膜间皮细胞connexin43 mRNA和蛋白质表达均较正常糖组显著下降(P<0.05)。渗透压对照组与正常糖组之间无明显差异(P>0.05)。结论高糖可抑制腹膜间皮细胞connexin43 mRNA和蛋白质表达,可能为腹膜透析(PD)中腹膜间皮层完整性损伤的机制之一。 Objective To investigate the effect of the high glucose concentration on the connexin43 expression in the cultured human peritoneal mesothelial cells(HPMC).Methods HPMCs isolated and cultured in vitro were divided into normal glucose group(5.5 mmol/L),high glucose group(140 mmol/L) and osmotic control group(5.5 mmol/L glucose group plus 24.5 mmol/L mannitol).At 24 h and 48 h of culture,the expression of connexin43 mRNA and protein was determined by Northern blot and Western blot,and the differences between the 3 groups were compared.Results HPMC in the normal glucose group expressed rich connexin43.The mRNA and protein expression of connexin 43 was reduced in high glucose group as compared to the normal group(P0.05).There was no evident difference in mRNA and protein expression of connexin43 between osmotic pressure group and the normal group(P0.05).Conclusion High glucose concentration can inhibit connexin43 expression in HPMC,which might contribute to the mechanism of the integral damage to the peritoneal mesothelium during prolonged peritoneal dialysis.
