大肠癌CD133(+)、Musashi-1(+)干细胞与临床病理学因素及MGMT、ERCC1、TS关系的研究 被引量:4

摘要 目的探讨大肠癌CD133(+)、Musashi-1(+)干细胞与临床病理因素及MGMT、ERCC1、TS的关系。方法应用免疫组织化学方法(SP法)检测42例大肠癌组织的CD133、CD138、Musashi-1、MGMT、ERCC1、TS蛋白,对各指标之间的相关关系及其与临床病理学因素之间的关系进行分析。结果①CD133与组织类型、Dukes分级、静脉侵袭、远处转移及毛细血管密度(MVD)等有关(P<0.05);CD138、Musachi-1与组织类型、浆膜侵犯、静脉侵袭、淋巴管侵袭、淋巴结转移、远处转移及MVD等有关(P<0.05),CD138并与神经纤维间隙侵袭(NI)有关(P<0.05);CD133与CD138的表达呈负相关(P<0.01),与Musashi-1的表达呈正相关(P<0.01)。②MG-MT、ERCC1与组织类型有关(P<0.05);TS与组织类型、Dukes分级、淋巴管侵袭、淋巴结转移有关(P<0.05);MGMT表达与ERCC1、TS表达呈负相关(P<0.01),ERCC1表达与TS表达呈正相关(P<0.01)。③CD133、Mu-sashi-1的表达均与MGMT的表达呈负相关(P<0.01),与TS、ERCC1的表达呈正相关(P<0.01)。结论①CD133(+)、Musashi-1(+)细胞在大肠癌的浸润与转移中起重要协同作用。②CD133(+)、Musashi-1(+)细胞与MGMT、TS、ERCC1耐药基因蛋白的表达具有密切关系,后者在大肠癌的侵袭中也具重要作用。 Objective To investigate the relationship between the colorectal cancer stem cells CD133(+) and Musashi-1(+) and the clinicopathological factors as well as proteins MGMT,ERCC1 and TS.Methods The immunohistochemical method(SP method) was used to detect stem cells CD133,CD138 and Musashi-1 and proteins MGMT,ERCC1 and TS in 42 cases of colorectal cancer tissues.The correlation between the various indicators as well as the relationship between these indicators and clinical pathological factors were analyzed.Results 1.CD133 was related to the histological typology,venous invasion,distant metastasis and capillary density(MVD)(P0.05);CD138 and Musachi-1 were related to the histological typology,serosal invasion,venous invasion,lymphatic vessel invasion,lymph node metastasis,distant metastasis and MVD(P0.05),CD138 was also related to the neural invasion(NI)(P0.05);CD133 was negatively correlated with the expression of CD138(P0.01),and was positively correlated with Musashi-1 expression(P0.01).2.MGMT and ERCC1 was related to the histological typology(P0.05);TS was related to histological typology,Dukes classification,lymphatic invasion and lymph node metastasis(P0.05);MGMT expression of ERCC1,TS expression was negatively correlated(P0.01),and ERCC1 expression was positively correlated with TS expression(P0.01).3.Either of the expressions of CD133 and Musashi-1 was negatively correlated with the expression of MGMT(P0.01),and was positively correlated with the expressions of TS and ERCC1(P0.01).Conclusion 1.Cells CD133(+) and Musashi-1(+) play an important synergetic role in the process of infiltration and metastasis of colorectal cancer.2.The stem cells CD133(+) and Musashi-1(+) are closely correlated with the expression of these resistance gene proteins(MGMT,TS and ERCC1),which also play an important role in the invasion of colorectal cancer.
