摘要
以牛皮杜鹃嫩茎段为外植体,应用均匀设计法筛选最适合的培养基。结果表明,最适合嫩茎段腋芽丛生芽苗的诱导培养基为1/2 MS+2-ip 3.50mg/L+IAA 0.02mg/L+GA3 1.00mg/L,诱导率可达96%;生根培养基为MS(改良)+IAA 0.10mg/L+NAA 0.07mg/L,生根率可达98%。以再生植株的茎节为材料进行快繁的结果表明,在30d的一个培养周期内,增殖倍数平均达60以上。通过试验,建立了牛皮杜鹃腋芽丛生芽苗及高频植株的再生体系。
To create a high frequency regeneration system from young stems of Rhododendron chrysanthum, suitable medium compositions for inducing multiple shoot clumps from axillary and rooting were screened through uniform design experiments. The results showed that 1/2 MS+2-ip 3.50 mg/L + IAA 0.02 mg/L + GA3 1,00 mg/L was effective for shoot clumps induction with the .frequency of 96%; MS (modified)+IAA 0.10 mg/L + NAA 0.07 mg/L was most suitable for plant rooting with the rate of 98%. The propagation results of stems from regenerated shoots showed that the proliferation rate was above 60 within 30 days culture period. The system of multiple shoot clumps from axillary and high frequency plant regeneration of R. chrysanthum was established.
