虎纹蛙皮肤组织cDNA文库的构建及抗菌肽基因tigerinin-HRs的克隆 被引量:1

摘要 运用SMART技术成功构建了虎纹蛙(Hoplobatrachus rugulosus)皮肤组织全长cDNA文库。根据蛙科动物抗菌肽的信号肽保守序列设计简并引物,以PCR法成功从文库中随机挑选的阳性克隆中筛选出2个抗菌肽cDNA序列,命名为tigerinin-HR1和tigerinin-HR2。这2个抗菌肽cDNA序列是首次从虎纹蛙皮肤组织中克隆出的抗菌肽编码基因。 A full length cDNA library of Hoplobatrachus rugulosus skin tissue was constructed with the SMART technique.A special primer was designed from a highly conserved domain of the signal peptide of antimicrobial peptide encoding cDNAs from ranid frogs.Two different cDNA sequences encoding precursor proteins named tigerinin-HR1 and tigerinin-HR2 were screened from the library by PCR.tigerinin-HR1 and tigerinin-HR2 were the first found encoding cDNA sequences of antimicrobial peptide from H.rugulosus.
