摘要
以长白舌唇兰和凹舌兰的茎尖为外植体,应用均匀设计法筛选最适合于原球茎和类原球茎诱导及类原球茎萌发为完整植株的培养基.结果表明,最适合长白舌唇兰原球茎和类原球茎诱导的培养基为N6+TDZ0.05 mg.L-1+NAA0.01 mg.L-1+KT0.50 mg.L-1,诱导率为92.5%;最适合凹舌兰原球茎和类原球茎诱导的培养基为N6+TDZ0.05 mg.L-1+KT0.50 mg.L-1,诱导率为98%;长白舌唇兰类原球茎萌发为完整植株的最佳培养基为N6+IAA0.01 mg.L-1,萌发率为96%;凹舌兰类原球茎萌发为完整植株的最佳培养基为N6+IAA0.01 mg.L-1+GA30.01 mg.L-1,萌发率为94.5%.以类原球茎的切片为材料进行类原球茎快繁的结果表明,在30~40 d的一个培养周期内,增殖倍数达100以上,成功建立了两种兰的高效快繁体系.同时对不同阶段培养材料的形态结构及超微结构的观察证明了两种兰的类原球茎发生发育过程.
The stems tip of Platanthera mandarinorum Rchb.f.var.cornu-bovis(Nevski) Kitag.and Coeloglossum viride(L.) Hartm were used as explants and suitable medium compositions for inducing protocorms and protocorm-like bodys,protocorm-like bodys germination were screened through uniform design experiments.The results showed that N6+TDZ0.05 mg·L^-1+NAA0.01 mg·L^-1+KT0.50 mg·L^-1 was most suitable for Platanthera mandarinorum with an induction rate of 92.5%;N6+TDZ0.05 mg·L^-1+KT0.50 mg·L^-1 was most suitable for Coeloglossum viride with an induction rate of 98%;N6+ IAA0.01 mg·L^-1 was most suitable for Platanthera mandarinorum with a germination rate of 96%;N6+IAA0.01 mg·L^-1+ GA30.01 mg·L^-1 was most suitable for Coeloglossum viride with a germination rate of 94.5%.Slices with one node were cut from induced protocorm-like bodys and cultured for propagation,and a 100-fold proliferation rate was achieved within 30~40 days.High effective micropropagation system of two Lilies has been successfully established.The obervation of morphostructure and ultrastructure proved the process of inducing and development for protocorm-like bodys of two Orchids.
