甘氨酰甘氨酸-铜(Ⅱ)-芳香氮碱配合物与DNA的作用 被引量:1

摘要 本文应用电子吸收光谱、溴化乙锭荧光光谱、粘度测定和琼脂糖凝胶电泳方法研究了甘氨酰甘氨酸-铜(Ⅱ)-芳香氮碱型配合物:[Cu(Gly-gly)(TATP)].2H2O(1)及[Cu(Gly-gly)(Phen)].3H2O(2)(Gly-Gly=甘氨酰甘氨酸,TATP=1,4,8,9-四氮三联苯,Phen=1,10-邻菲咯啉)与DNA之间的相互作用。结果表明,这些配合物通过插入作用与DNA结合,结合能力大小为:配合物1>2;在维生素C存在下对pBR322 DNA具有显著的氧化断裂作用;活性大小为:配合物1>2。 The interaction of copper( Ⅱ )complexes: [ Cu(Gly-gly) (TATP) ] · 2H2O( 1 ) ) and [ Cu(Gly-gly) (Phen) ] ·3H2O(2) ( (Gly-gly = Glycyl-glycine, TATP = 1,4,8,9-tetmnitmgen Phen = 1,10-phenanthroline )with DNA has been investigated by electronic absorption spectroscopy,ethidium bromide (EB)fluorescence spectroscopy, viscosity and agarose gel eleetrophoresis measurements. The results indicated that the complexes could bind to DNA by interealatiive mode, and cleave pBR322 DNA in the presence of vitamin C with the order of the binding ability and cleavage activity:complex 1 〉 2.
