高效液相色谱法同时测定蝙蝠葛酚性碱片中蝙蝠葛碱和蝙蝠葛苏林碱含量

Simultaneous determination of dauricine and daurisoline in phenolic alkaloids of menispermum dauricum tablets by HPLC

目的建立同时测定蝙蝠葛酚性碱片中蝙蝠葛碱和蝙蝠葛苏林碱含量的HPLC法。方法采用Diamonsil C18色谱柱（4．6mm×200mm，5μm）；流动相：乙腈-4％冰醋酸-三乙胺（40：220：1．5）；流速：1．0mL·min^-1，检测波长：282nm；柱温：30℃。结果蝙蝠葛碱和蝙蝠葛苏林碱的线性范围分别为9．28-92．8μg·mL^-1（R=0．9999）和2．8～28．0μg·mL^-1（R=0．9999）。平均回收率分别为97．7％（RSD=1．1％）和99．4X（RSD=2．0％）。结论本方法简便、准确、灵敏度高、重现性好，可作为蝙蝠葛酚性碱片的质量控制方法之一。

Objective To establish an HPLC method for the simultaneous determination of dauricine and daurisoline in phenolic alkaloids of menisperrnum dauricum tablets. Methods The samples was separated on a Diamonsil C18 column （4. 6mm×200mm,5μm） at 30℃. The mobile phase consisted of acetonitrile-4% glacial acetic acid-triethytamine （40 ： 220 ： 1.5）. The flow rate was 1.0 mL· min^-1 and the detective wavelength was 282 nm. Results The calibration curves of dauricine showed good linearity over 9.28--92. 8μg·mL^-1 （R=0. 999 9）. The average recovery was 99.4%, and RSD was 2.0%. The calibration curves of daurisoline showed good linearity over 2.8--28. μg·mL^-1 （R=0. 999 9）. The average recovery was 97. 7%, and RSD was 1.1%. Conclusion The method is simple, accurate and sensitive with good reproducibility. It can be used to control the quality of phenolic alkaloids of menispermum dauricum tablets.
