摘要
目的通过对槐定碱致痫大鼠脑内Caspase3及热休克蛋白-70(hsp-70)含量的测定,探讨细辛脑注射液抗癫痫作用的机制。方法随机将42只大鼠分为6组,空白组、模型组、卡马西平阳性对照组及细辛脑低剂量组、中剂量组和高剂量组。通过免疫组化学法和Nissle染色法,研究细辛脑注射液对Caspase3和hsp-70含量的影响。结果研究表明与模型组比较,细辛脑注射液中、高剂量组可显著减轻大鼠癫痫发作(P<0.05),可使槐定碱致痫大鼠的脑组织中减少Caspase3释放,降低hsp-70在大鼠脑内的表达。结论细辛脑注射液具有良好的抗癫痫效果,可减轻大鼠癫痫发作时对脑组织的损伤。
Objective To explore the anti-epileptic effect and its mechanism of Asarone injection on Caspase 3 and heat shock protein-70(hsp-70)in sophoridine-induced epileptic rats.Methods Forty-two rats were randomly divided into six groups:Blank group,sophoridine-induced epilepsy model group,carbamazepine group,Asarone injection low dosage group,Asarone injection medium dosage group and Asarone injection high dosage group.The content of Caspase 3 and HSP-70 were detected by immunohistochemistry and Nissle staining.Results Compared with model group,the seizure were reduced significantly and the content of Caspase 3 and HSP-70 in brain tissue were decreased in Asarone high dosage group(P 0.05).Conclusion Asarone injection had anti-epileptic effect,which could play a role in preventing seizure of rats effectively.
