补中益气汤对脾虚型复发性口腔溃疡小鼠IL-2 IFN-γ表达的影响

Influence of Buzhongyiqitang over the spleen deficiency type recurrent oral ulcers mice IL-2 IFN-γindex

目的:通过观察实验小鼠口腔溃疡情况及检测外周血血清中IL-2、IFN-γ水平,来评价补中益气汤对实验性脾虚型复发性口腔溃疡小鼠的治疗效果,探讨其作用机制,从而进一步为临床应用提供理论依据。方法:①先用限制饮食和过度劳倦法建立小鼠脾虚模型,然后用抗代谢化疗药甲氨喋呤腹腔注射,3种方法联合应用复制小鼠脾虚型口腔溃疡模型;②检测各组小鼠外周血中IL-2、IFN-γ水平。并取正常及有损伤的口腔黏膜组织,做病理检查;③再将剩余模型小鼠随机分为补中益气汤治疗组和生理盐水对照组,观察每组小鼠一般症状体征及口腔溃疡变化情况,检测IL-2、IFN-γ含量。结果:模型小鼠口腔黏膜可见黏膜有上皮层坏死、脱落,表面覆盖大量中性粒细胞和纤维性渗出物,黏膜伴见炎症细胞浸润;模型组血清IL-2、IFN-γ含量水平显著下降(P<0.01);补中益气汤治疗组一般情况恢复快,口腔溃疡愈合快,复发少,IL-2、IFN-γ水平显著升高。结论:①补中益气汤可以显著改善脾虚症状,促进溃疡愈合,减轻症状,减少溃疡复发;②补中益气汤可以升高IL-2、IFN-γ,增强机体免疫功能;③脾虚型复发性口腔溃疡的发生与免疫功能下降有着重要的关系。

Objective：By observing the experimental mice oral ulcer and peripheral blood serum IL-2,IFN-Γ level,to evaluate Buzhongyiqitang therapeutic effect in the spleen deficiency type recurrent oral ulcers mice,discuss the effect mechanism,in order to provide a theoretical basis for clinical application. Methods：（1） Use restricted diet and excessive fatigue way to establish a mice model of spleen deficiency,on this basis,inject with antimetabolite chemotherapy drug-intraperitoneal methotrexate,once a day,continuous for five days. Joint use the three methods to reproduce the model of spleen deficiency type recurrent oral ulcers mice successfully. （2） Detect normal mice and model mice peripheral blood of IL-2,IFN-γ level.Check normal and damaged oral mucosa,and do pathological examination. （3） Then divide remaining model mice into Buzhongyiqitang treatment group and saline treatment comparison group randomly,both groups give normal diet,observing general symptoms and the oral ulcers change of mice in each group. After 10 days treatment,test IL-2,IFN-Γ content.Results：Observation of oral mucosa pathological organization showed that：the normal group mice＇s oral mucosa is smooth,no significant inflammatory cell infiltration. Model group mice＇s Oral mucosal ulcers can see-there is necrosis of mucosa epithelium,detached,surface coverage of a large number of neutrophils and fibrous exudation,mucous membrane with infiltration of inflammatory cells. Model mice serum IL-2,IFN-γ levels decreased significantly （P0.01） than normal mice. Compared with saline treatment comparison group,generally,Buzhongyiqitang treatment group resumption and healing oral ulcers faster,recurrent lower,the level of IL-2,IFN-γ have increased significantly.Conclusion：①Buzhongyiqitang can improve spleen deficiency symptoms significantly,promote ulcer healing and reduce symptoms,reduce ulcer recurrence.②Buzhongyiqitang can increase IL-2,IFN-γ,enhance immune function.③The occurrence of spleen deficiency type recurrent oral ulcer has important relationship with decreased immune function.
