标本放置时间对胰岛素测定的影响

摘要 目的探讨血清标本在4℃放置不同时间对化学发光检测血清胰岛素浓度的影响。方法用真空分离胶采血管抽取21例血标本,冷冻离心分离血清,采用全自动化学发光仪即时检测胰岛素浓度,将标本于4℃分别放置1小时、3小时和24小时后重复检测,并与即时检测结果相比较。结果标本采集后即时检测与4℃下放置1小时、3小时、24小时后检测胰岛素浓度分别为(6.88±4.38)mIU/L、(6.87±4.44)mIU/L、(7.05±4.68)mIU/L、(7.57±4.60)mIU/L,经配对t检验,血清标本在4℃放置1小时和3小时所测结果与即时测定结果无统计学差异,而放置24小时后所测结果与即时测定结果有统计学差异(P=0.009)。结论标本放置时间对胰岛素测定有影响,血清胰岛素在4℃条件下不能放置24小时以上。 Objective To study the influence of serum sample storage time on the contents of serum insulin detected by chemiluminesence. Methods 21 serum samples were selected, the contents of serum insulin were detected by chemiluminesence immediately. Measurement of insulin of all serum samples which were stored in 4℃ were repeated in 1 hour, 3 hours and 24 hours later respectively. Results The contents of serum insulin at 0 hour and 1 hour, 3 hours, 24 hours were (6.88 ± 4.38) mlU/L, (6.87 ± 4.44) mIU/L, (7.05 ± 4.68) mIU/L, (7.57 ± 4.60) mIU/L respectively. There were no significant differences among the levele of serum insulin detected at 0 hour, 1 hour and 3 hours, while there was significant difference between contents of serum insulin detected at 0 hour and 24 hours. Conclusion The duration of the serum sample storage time markedly influences the accuracy of the determination of insulin. All determination should be finished as soon as possible. Serum insulin at 4 ℃ can not be placed more than 24 hours.
