摘要
目的研究蝙蝠葛酚性碱(PAMD)对BXPC-3裸鼠抑瘤作用及对肿瘤新生血管内皮因子(VEGF)表达的影响,并进一步探讨其作用机制。方法建立BXPC-3裸鼠的动物模型,检测蝙蝠葛酚性碱对转移瘤的抑瘤率,采用免疫组织化学方法检测各组BXPC-3小鼠肿瘤组织VEGF的表达水平。结果蝙蝠葛酚性碱对BXPC-3胰腺癌裸鼠瘤组织具有一定的抑制作用,与模型对照组比较具有统计学意义(P<0.01)。PAMD各剂量均能明显降低BXPC-3胰腺癌裸鼠瘤组织VEGF的表达水平。结论蝙蝠葛酚性碱对BXPC-3裸鼠胰腺癌具有明显的抑制作用。其机制可能是通过改善裸鼠免疫功能,抑制肿瘤新生血管形成来实现的。
Objective To explore the effects of phenolic alkaloids from menispermum dauricum(PAMD)on tumor inhibitory and expression of basic fibroblast growth factor( VEGF) in BXPC 3 nude mlee and to further inves- tigate its protective mechanism for pancreatic carcinoma. Methods The metastatic tumor model of nude mice was established with BXPC-3 cell line. After PAMD treatment, the inhibition rate of tumor proliferation was detected and immunohistochemistry was carried out to confirm the expression of VEGF in nude mice. Results PAMD could inhibit the tumor growth of tumor-bearing mice of BXPC-3 cell line(P〈0. 01, vs control group). Meanwhile, PAMD of different doses could inhibit the expression of VEGF in tumor tissue of nude mice. Conclusion PAMD shows conspicuous inhibitory action on the tumor growth of nude mice,which possibly due to its potency to improve immune function and inhibit tumor angiogenesis in nude mice.
