摘要
目的:探讨吗啡预处理对大鼠脑缺血再灌注损伤后脑含水量及脑脊液S100β的影响。方法:Pusinelli方法建立四动脉阻断全脑缺血模型:成年雄性大鼠36只,随机分为假手术组(n=12)、脑缺血再灌注组(n=12)、吗啡预处理组(n=12)。脑缺血8min,再灌注48h后,每组取6只大鼠断头取脑制作石蜡切片,HE染色观察海马区组织病理学改变;另取剩余6只大鼠于枕骨大孔抽脑脊液后立即断头取脑,干湿称重法测量脑含水量,酶联免疫吸附法检测脑脊液中S100β蛋白含量。结果:吗啡预处理组细胞受损的病理学改变显著轻于脑缺血再灌注组。吗啡预处理组脑含水量及S100β蛋白含量明显低于脑缺血再灌注组(P<0.05或P<0.01)。结论:吗啡预处理可减轻缺血性脑损伤,提高脑缺血耐受性。
Objective:To investigate effects of morphine preconditioning on brain water content and S100beta of cerebrospinal fluid(CSF) in rats with cerebral ischemia-reperfusion injury.Methods:36 male rats were randomed divided into three groups:sham-operated group(n=12);cerebral ischemia-reperfusion group(n=12);morphine preconditioning group(n=12).Global cerebral ischemia model were produced by four-vessel occlusion as described by Pusinelli.Ischemia models were made to occlude carotid artery for 8 min,then which were loosened to reperfusion.At the end of 48 h reperfusion,Hippocampus tissue were obtained for HE staining to detect pathological changes and brain were obtained to weigh and CSF were obtained to detect S100beta protein.Results:Pathological changes of neuron in morphine preconditioning group were significantly lighter than that in cerebral ischemia-reperfusion group.Compared with that in cerebral ischemia-reperfusion group,brain water content and values of S100beta in morphine preconditioning group decreased significantly(P〈0.05 or P 〈0.01).Conclusion:Morphine preconditioning can reduce ischemic brain injury and improve cerebral ischemic tolerance.
