摘要
利用前期获得并经基因工程改造的一株高效几丁质酶降解菌进行发酵产酶法降解几丁质胶体与膜分离相耦合制备几丁寡糖的中试研究.经过中试发酵工艺、酶反应工艺、分离纯化工艺的研究,最终确定了一条高效制备几丁寡糖的中试规模工艺流程,该流程中反应分离一体化,过程实现连续操作,降解酶可以连续使用,提高了收率,降低了成本,有利于实现产业化.通过该流程制备的几丁寡糖混合物含几丁2、3、4糖分别为71.3%、23.5%、2.4%.
Chitin oligosaccharides was produced by chitinases digestion of chitin colloid coupling with membrane separation. The chitinases was produced by fermentation through wild (aeromonas caviae CB101 ) bacteria changed by our group. A method suitable for industrialization was suggested through detail fermentation,enzyme digestion, and separation study. The character of this method is digestion and separation can be operated at same time in series. The purity of N-Acetylchitooligosaccharides (dimer, trimer,tetram- er) produced by this method is 71.3%, 23.5%,2.4%. Chitinases can be used more effectively and with high yield and low cost in favor the development of industrialization.
