摘要
应用RT-PCR技术对贵州省不同时期不同禽源NDV分离株(肉鸡源P1株与BY株、蛋鸡源H2株与FW株、七彩山鸡源N98株、越南斗鸡源DQ株、鸵鸟源TN株及鸽源GZ株)F蛋白基因进行扩增、克隆和测序分析,结果表明:8株NDV贵州分离株F基因长度均为1 662 bp,可编码553个氨基酸;NDV贵州分离株间核苷酸同源性为84.0%-99.7%,氨基酸同源性为87.5%-99.3%;与国内外NDV参考毒株(LaSota株、B1株、F48E9株、CH2 000株和TW2 000株)的核苷酸同源性为84.2%-98.9%,氨基酸同源性为87.2%-98.6%;GZ株和TN株为基因II型,DQ株、N98株、P1株和H2株为基因VII型(均为VIId亚型),BY株和FW株为基因IX型。表明,不同时期NDV贵州分离株的基因型不同,近年流行的ND疫情主要是由基因VII型NDV引起,与国内其他省市ND流行趋势基本一致。
The fusion protein gene(F gene) of eight isolates of Newcastle disease virus(NDV),which is isolated from different avian in different years in Guizhou province,were amplified using RT-PCR technique,then were cloned respectively into the pMD18-T vector and sequenced,and these F gene were analyzed with reference NDV stains in GenBank.The results showed that F gene of all isolates are 1662 bp encoding 553 amino acids,and the sequence identity matrix indicated that the homology of nucleotide sequence of these isolates are 84.0%~99.7% and that of amino acid sequence are 87.5%~99.3%,and they are respectively 84.2%~98.9% and 87.2%~98.6% compared with that of Lasota,B1,F48E9,CH2000 and TW2000.The phylogenetic analysis showed that strain DQ,N98,P1 and H2 belonged to VII genotype,strain BY and FW to IX genotype,and strain GZ and TN to II genotype.The results revealed that the genotype of NDV had transformed in different years,and VII genotype is the main genotype of NDV occurred in Guizhou province now,basically as the same trend of ND in the other provinces.
