摘要
目的建立泰山女儿茶中总黄酮和多糖的含量测定方法。方法以分光光度法测定总黄酮含量,用苯酚-硫酸法测定多糖含量。结果总黄酮含量测定最佳检测波长510 nm,芦丁质量浓度在8~48μ.mL-1范围内与吸光度呈良好线性关系(r=0.9986),平均回收率99.03%(RSD=1.72%,n=5);多糖含量测定的检测波长490 nm,葡萄糖质量浓度在5~50μ.mL-1范围内与吸光度呈良好线性关系(r=0.9964),平均回收率99.26%(RSD=1.57%,n=4)。结论本方法简便易行,结果稳定可靠,可用于泰山女儿茶的质量控制。
Objective to establish the methods for determination of total flavonoids and polysaccharides in Nv'er Tea from Taishan.Methods the content of the total flavonoids was determined by spectrophotometry,and the polysaccharide content was measured with the phenol-sulphuric acid method.Results the optimal detective wavelength was 510 nm for the total flavonoids,and satisfactroy linear relationship with the absorbability was achieved when the mass concentration of rutin was within a range of 8~48 u·mL-1(r=0.9986),the average recovery rate was 99.03%(RSD=1.72%,n=5).The optimal detective wavelength was 490 nm for the polysaccharides,there was good linear relationship between the absorbability and the content of glucose in a range of 5~50 u·mL-1(r=0.9964) with an average recovery of 99.26%(RSD=1.57,n=4).Conclusion this method is sensitive,simple and accurate,and it can be used for the quality control of Nv'er Tea from Taishan.
