腹部开放伤合并海水浸泡后致肠功能障碍半胱氨酸蛋白酶-3的表达与细胞凋亡的关系 被引量:4

摘要 目的研究腹部开放伤合并海水浸泡所致肠功能障碍发生过程中小肠上皮细胞的半胱氨酸蛋白酶(Caspase)的表达与细胞凋亡的关系。方法原位末端标记(TUNEL)法检测大鼠腹部致伤后不浸泡、0.9%氯化钠溶液浸泡、海水浸泡结束后细胞凋亡的发生及变化特点。免疫组化检测各组凋亡相关蛋白Caspase-3表达变化。结果正常大鼠小肠上皮散在TUNEL阳性细胞,Caspase-3阳性细胞主要在小肠上皮绒毛处呈中等阳性表达。腹部致伤后不浸泡及0.9%氯化钠溶液浸泡组TUNEL阳性细胞较对照组增多,而Caspase-3表达亦较对照组增高。海水浸泡组的TUNEL阳性细胞及Caspase-3表达较0.9%氯化钠溶液浸泡组及不浸泡组明显增多。结论在大鼠腹部开放伤合并海水浸泡导致的肠功能障碍无论从形态学上及凋亡蛋白的表达上均较不浸泡和0.9%氯化钠溶液浸泡严重,大量肠细胞凋亡的发生可能是海战伤中早期出现肠功能障碍的主要原因。 Objective To study the correlation between intestinal epithelial apoptosis and expression of caspase-3 in intestinal dysfunction caused by the open abdominal wound with seawater immersion.Methods The in situ terminal deoxymudeotityl transferase mediated dUTP nick end labeling (TUNEL) was used to detect the occurrence and change characteristics of apoptosis in rats with abdominal wound treated respectively with nom-immersion,physiological saline immersion,and seawater immersion.And immunohistochemical technique was taken to detect the expression features of caspase-3.Results In normal rats intestinal epithelial scattered TUNEL-positive cells and caspase-3-positive cells mainly showed moderate positive expression in the small intestine epithelial villi.In rates with abdominal wound of the non-immersion and physiological saline immersion group TUNEL-positive cells and caspase-3 expression increased as compared with the control group.The TUNEL-positive cells and the caspase-3 expression in seawater immersion group increased significantly than those in the physiological saline group and non-immersion group.Conclusion The rats with intestinal dysfunction caused by open abdominal wound treated with seawater immersion are more serious than those treated with non-immersion and fresh water immersion in terms of morphology and expression of apoptosis.The apoptosis of a large amount of intestinal cells may be the main reasons for the early emergence of intestinal dysfunction in sea battle injuries.
