摘要
比较三氯乙酸/丙酮沉淀法、酚柚提法两种方法对锥栗叶片总蛋白的提取效果。结果表明:酚抽提法获得的蛋白质产率高、纯度高;且其提取蛋白质所得电泳图谱背景浅、拖尾轻、蛋白点分散且性状规则,适合锥栗叶片蛋白质的提取。试验为锥栗胚胎发育期蛋白质组学研究提供参考,也对其他干扰物质含量高的植物组织蛋白提取有一定的参考价值。
The effect of the general protein extraction ofCastanen henryi Rehd & Wils leaves between trichloroaeetic acid- acetone precipitation method(TCA method) and phenol isolation method(phenol method) has been compared. The results were as follows: the yield ratio and purity of the protain by phenol method were better than those by TCA method; phenol isolation method could obtain a low-background graph with few blurred and smearing spots; besides, the spots scatterd and had regular shape, and thus this method is optimal for extracting leaves protein from Castanen henryi Rehd & Wils. As a result, the test results can provide reference for the proteomics research of Castanen hemyi Rehd & Wils embryo during the developing period and have significant value for guiding the protein extraction of many other plants containing various interfering elements.
