摘要
利用荧光光谱法、紫外-可见吸收光谱法、圆二色谱法、凝胶电泳法对铬天青S(CAS)与核酸的作用机理进行了探讨,当ct-DNA浓度增加时,铬天青S的荧光光谱呈荧光增强趋势,紫外-可见光谱呈下降趋势,并使ct-DNA圆二色谱正负峰的吸收强度减小。琼脂糖凝胶电泳实验表明,铬天青S-DNA在泳道中清晰可见。将所得结果与溴化乙锭(EB)-DNA体系性质进行对比,得出铬天青S与DNA有发生类似于溴化乙锭的嵌插作用的结论。
The interactions between chrome azurol S (CAS) and DNA were studied by fluorescence spectroscopy,UV-absorption spectroscopy,circular dichroism (CD) spectroscopy and agarose gel electrophoresis method.With an increasing DNA concentration,the fluorescence intensity of CAS increased accordingly.The absorbance of CAS on UV spectra demonstrated a hypochromic effect with an increasing DNA concentration.The CD spectra of DNA indicated a great change both in terms of negative and positive absorption peaks with CAS added.With regard to the agarose gel electrophoresis analysis,the luminescence intensity of CAS-DNA system could be seen clearly.By comparing the above study results with EB-DNA system,we came to conclusion that chrome azurol S and ethidium bromide displayed the same modes when they interacted with DNA.
