异丙酚抑制BALB/c小鼠结肠癌细胞株Colon26肺转移的实验研究 被引量:1

摘要 目的观察异丙酚对BACL/c小鼠结肠癌细胞株Colon26肺转移的影响并探讨其机制。方法结肠癌细胞株Colon26从BACL/c小鼠尾静脉注射,建立鼠结肠癌肺转移模型;采用随机数字法将30只小鼠随机分为3组,每组10只,24h后经腹腔注射:生理盐水5mL/kg(A组),英脱利匹特5mL/kg(B组),异丙酚50mg/kg(C组),1次/d,2周后观察肺转移情况,计算肺转移结节数,以乳酸脱氢酶同功酶(LDH)释放法测定自然杀伤细胞(NK)活性;高效液相芯片技术检测外周血中IFN,小鼠IL-12的含量。结果肺转移结节数3组分别为A组12.6±2.3,B组10.9±2.1,C组5.2±1.2,C组肺转移结节数明显低于A组(P<0.05),B组与A组差异无统计学意义;C组NK细胞活性明显高于其他两组;血清中IFN-α平均含量分别为A组(25.31±9.52)pg/mL、B组(32.67±7.64)pg/mL、C组(85.62±10.27)pg/mL,C组含量明显高于其他两组(P<0.05);IL-12平均含量分别为A组(15.76±5.56)pg/mL、B组(18.28±8.13)pg/mL、C组(45.42±6.18)pg/mL,C组与其他两组比较差异有统计学意义(P<0.05)。结论异丙酚能通过增加NK细胞活性,促进IFN-α、IL-12分泌,从而抑制BALB/c小鼠结肠癌肺转移。
