摘要
分离和鉴定了采自中国南海硇州岛软珊瑚Sinularia Bassica样本中新的嘧啶类化合物。样本用工业酒精提取,提取物溶液经乙酸乙酯萃取后,萃取物经硅胶柱层析,以极性不断增大的溶剂体系(石油醚-乙酸乙酯、氯仿-甲醇)梯度洗脱,再经过HPLC分离,得到2,4(1H,3H)-嘧啶二酮、5-甲基-2,4(1H,3H)-嘧啶二酮、1,3-二甲基-2,4(1H,3H)-嘧啶二酮和5-甲氧基-1,3-二甲基-2,4(1H,3H)-嘧啶二酮4种新的嘧啶类化合物,其结构通过红外光谱、1HNMR和13C NMR等光谱数据分析确定。
Isolation and identification of new pyrimidine analogues from soft coral Sinularia Bassica collected from the Naozhou Island of the South China Sea were investigated. Dry Sinularia Bassica was extracted with industrial alcohol. The extract was concentrated under reduced pressure condition and the aqueous residue was partitioned with ethyl acetate. The fraction of ethyl acetate was chromatographed with the silica gel of 50 - 75 μm (200 - 300 mesh) by increasing polarity of solvents from petroleum ether- ethyl acetate through CHC13 to CHC13 -CH3OH mixtures. Four new pyrimidine analogues from the fraction were isolated by the HPLC, and their structures were identified as 2, 4 ( 1 H, 3 H) -pyrimidinedione, 2, 4 ( 1 H, 3 H) -pyrimidinedione, 5-methyl, 2, 4 ( 1 H, 3 H ) -pyrimidinedione, 1, 3-dinemethyl and 2, 4( 1H, 3H) -pyrimidinedione, 5-methoxy-1, 3-dimethyl based on the analysis of infrared (IR) and 1H and 13C nuclear megnetic resoance ( including 1D and 2D NMR) spectra.
