摘要
目的建立蚕蛹蛋白氨基酸含量的测定方法。方法采用柱前衍生反相高效液相色谱法。结果在给定的色谱条件下,18种氨基酸在35 min内获得了较好的分离,在一定浓度范围内,氨基酸的峰面积与浓度的线性相关系数在0.990 6-0.999 8之间,相对标准偏差为2.02%-5.45%(n=5)。并测定了蚕蛹蛋白中氨基酸的组成和含量。结论该方法准确可靠、重复性好,可用于蚕蛹蛋白中氨基酸含量的测定。
Objective To establish a method for determination of amino acids in proteins of silk worm.Methods A reversed-phase high-performance liquid chromatographic method for the determination of amino acids was described.Results The mixture of 18 kinds of amino acids was characterized by HPLC in 36 min.The response was linear for all amino acids with a correlation coefficient of 0.990 6-0.999 8,method precisions(n=5) for the different amino acids were between 2.02%-5.45%(relative standard deviation,RSD).Using this method,18 kinds of amino acids in protein of silk worm were determined.Conclusion The method is accurate,reliable and repeatable for the determination of amino acids in proteins of silk worm.
