PLGA-蒿属花粉纳米疫苗对小鼠过敏性哮喘治疗作用及机制的实验研究

摘要 目的研究以聚乳酸-羟基乙酸共聚物[poly(D,L-lactic-co-glycolic)acid,PLGA]为佐剂制备的蒿属花粉纳米疫苗对小鼠过敏性哮喘模型气道炎症治疗的疗效,并探讨其免疫治疗机制。方法40只小鼠随机分为5组:对照组、模型组、空白粒子治疗组(治疗A组)、蒿属花粉粗浸液治疗组(治疗B组)、PLGA-蒿属花粉治疗组(治疗C组)。用蒿属花粉建立哮喘模型。对照组用生理盐水代替致敏。ELISA法检测各组小鼠支气管肺泡灌洗液(BALF)和脾细胞培养上清液(SCCS)中IL-4,IFN-γ的含量;收集BALF进行总细胞计数和嗜酸性粒细胞计数;HE染色观察各组小鼠肺组织病理变化。结果肺部组织学和BALF中细胞计数结果显示,与模型组相比,B、C组的肺部嗜酸性粒细胞浸润显著减轻,BALF中总细胞和嗜酸性细胞显著减少;细胞因子的测定结果显示,在BALF中,模型组的IL-4和IFN-γ分别为(88.71±6.82)pg/ml和(24.14±2.91)pg/ml,与之相比,蒿属花粉粗浸液治疗组(40.07±2.42)pg/ml和PLGA-蒿属花粉治疗组(38.31±6.28)pg/ml的IL-4显著降低(P<0.01);PLGA-蒿属花粉治疗组(63.69±8.0)pg/ml的IFN-γ显著升高(P<0.01);在SCCS中,模型组的IL-4和IFN-γ分别为(81.02±12.5)pg/ml和(34.12±4.64)pg/ml,与之相比,蒿属花粉粗浸液治疗组(58.91±9.10)pg/ml和PLGA-蒿属花粉治疗组(41.30±4.75)pg/ml的IL-4分泌显著减少(P<0.01);PLGA-蒿属花粉治疗组(82.50±5.63)pg/ml的IFN-γ显著增高(P<0.01)。结论PLGA-蒿属花粉纳米疫苗免疫治疗小鼠哮喘有良好疗效。其机制可能与调节Th1/Th2平衡有关。
