养肝和血方对初老雌性大鼠卵巢Bcl-2和Bax表达的影响 被引量:2

Impact of Yanggan Hexue Fang on the Expression of Ovary Bcl-2 and Bax in Initial Aging Female Rats

摘要 目的探讨养肝和血方对初老雌性大鼠卵巢Bcl-2和Bax表达的影响,并以此论证在围绝经前期从肝论治的思想。方法通过阴道细胞学观察,选择10~11个月龄自然衰老雌性大鼠30只,随机分为模型对照组、六味地黄汤组、养肝和血方组,另选4个月龄的雌性大鼠10只为青年对照组。灌胃3周后,采用免疫印迹法(Western-blot法)检测各组大鼠卵巢细胞Bcl-2和Bax蛋白的表达。结果与模型对照组、六味地黄汤组比较,养肝和血方组卵巢细胞Bcl-2蛋白表达明显增高,Bax蛋白表达明显降低,Bcl-2/Bax比值增高,差异有统计学意义(P<0.05)。结论养肝和血方有抑制初老雌性大鼠卵巢细胞凋亡作用,可能是养肝和血方延缓卵巢衰老的机制之一。 Objective To explore the impact of yanggan hexue fang(the formula for nourishing the liver and harmonizing blood)on the expression of ovary Bacl-2 and Bax in the initial aging female rats so as to discuss the idea of liver therapy at premenopausal stage. Methods Virginal cytological observation was adopted. 30 natural aging female rats of 10 to 11 months old were selected and randomized into model control group,liu wei di huang tang group,and yanggan hexue fang group. Additionally,the other 10 female rats of 4 months old were selected in youth control group. 3 weeks after gastric infusion,western-blot method was used to determine the expression of the ovary cellular Bcl-2 and Bax in rats of each group. Results Compared with model control group and liu wei di huang tang group,the expression of ovary cellular Bcl-2 protein in yangguan hexue fang group increased apparently and that of Bax protein decreased apparently. The ratio of Bcl-2/Bax increased,indicating the statistical significant difference(P0.05). Conclusion Yanggan hexue fang inhibits ovary apoptosis in the initial aging female rats,which may be one of the mechanisms of the treatment to delay the ovary aging.
