局部应用卡铂抗肿瘤实验研究

目的探讨化疗不同给药途径的抗癌作用，为局部应用卡铂化疗提供依据。方法用Ｓ－１８０实体瘤小鼠模型研究比较静脉和瘤体注射卡铂６０ｍｇ／ｋｇ的药代动力学。于注药后３０分钟、９０分钟、２４０分钟分别取血，用高效液相色谱法测定卡铂浓度。结果局部瘤体给药组血浓度是１５．０３μｇ／ｍｌ、１７．０６μｇ／ｍｌ、１４．０３μｇ／ｍｌ。静脉注射组分别是４８．４５μｇ／ｍｌ、３９．３１μｇ／ｍｌ、３４．８５μｇ／ｍｌ。局部注射组抑瘤率明显高于静脉注射组（Ｐ＜０．０５）。结论实验证明，局部化疗能提高瘤体内药物浓度，提高杀瘤细胞作用，降低了循环药物浓度，从而减轻全身毒副作用。因此。

Objective To provide evidence of local chemotherapy for carboplatin,and anticancer effects of it by different routes of administration.Methods Pharmacokinectic comparison of intratumor (iT) and intravenous (iv) carboplatin 60mg/kg chemotherapy was done on mice S-180 tumor moder.High performance liguid chromatographay was used to measure the concentration of carboplatin after administration 30 minutes、90minutes、240 minutes,respectively.Results The blood drug concentration of “iT group”was 15.03μg/ml,17.06μg/ml,14.03μg/ml and that of “iv group” was 48.4 μg/ml,39.31μg/ml,34 85μg/ml,respectively.The antitumor rate was significant greater for “iT group”than“iv group”(P<0.05).Conclusions Local chemotherapy provide higher drug concentration within the tumor.decreased drug concentration of circulation and less body toxicity.Therefore local chemotherapy was an efficiency method for treating malignant tumor.
