摘要
本文探讨不同浓度的透明质酸(HA)对小鼠卵细胞的影响.方法:5~6周B_6C_3F_1雌鼠经孕马血清(PMSG)和人绒毛膜促性腺激素(hCG)促排卵,取同系12~16周雄鼠精子.实验1组,卵细胞团直接置不同浓度HA中,孵育不同时间后行体外授精;实验2组,卵细胞团先经透明质酸酶消化,然后置不同浓度HA中孵育不同时间后行体外授精;实验3组、卵细胞团不经HA处理,孵育不同时间后行体外授精.观察各组受精率及胚胎发育情况.结果:实验1组和实验3组受精率及胚胎发育情况优于对照组(P<0.01和P<0.05).实验2组受精率及胚胎发育情况无明显改善(P>0.05).结论:用HA孵育鼠卵细胞对受精及胚胎发育有利,内源性HA合成可能起一定作用,外源性HA浓度有待进一步探讨.
To evaluate the effects of exogenous hyaluronic acid (HA) of various concentration on mouse oocytes and fertilization rate. 5~6 week-old B6C3F female mice were superovulated by injection pregnant mare's serum gonadotrophin (PMSG ) followed by human chorinic gonadotrophin (hCG) given by intraperitoneally. Spermtozoa were obtained from the 12~16 week - old B5C3F, male mice. Experiment 1 :The oocytes with cumulus cells(CCs) mass were incubated directly with HA of various concentration and inseminated in vitro. Experiment 2; The oocytes with CCs were digested with hyaluronicase and then incubated with HA of various concentration and were insem-inated. Experiment 3: The oocytes with CCs were incubated without HA.and then inseminated. The fertilization rate and development into hatched in vitro in experiment 1 and 3 were significant-ly higher than those in the controls (P<0. 01 and f<0. 05) .while in experiment 2 were no sig-nificantly higher than those in control (.P>0. 05). It is beneficial to fertilization rate and embryos that hyaluronic acid incubated mouse oocytes -cell. Synthesis of endogenous HA could play a role in the process. The concentration of exogenous HA would be studied in future.
